amITT population consists of all subjects in the ITT population who received at least 1 dose of study medication and had at least 1 baseline and postbaseline efficacy assessment.
Inclusion criteria1,2
ULTIMATE I and II primary, select secondary, and select tertiary endpoints. Bold indicates statistically significant difference favoring BRIUMVI. Italics indicates endpoints not considered statistically significant between treatment groups. The tertiary endpoints below were not formally tested for significance.
Week 96 | • The number of new or enlarging T2 hyperintense lesions by Week 96 • Time to CDP for at least 12 weeks | • Time to CDP for at least 24 weeks (descriptive analysis) • Time to CDI for at least 12 weeks (descriptive analysis) • Time to CDI for at least 24 weeks (descriptive analysis) |
ARR, annualized relapse rate; CDI, confirmed disability improvement; CDP, confirmed disability progression; DMT, disease-modifying therapy; EDSS, Expanded Disability Status Scale; Gd+, gadolinium-enhancing; MRI, magnetic resonance imaging; ITT, intention-to-treat; mITT, modified intention-to-treat; RMS, relapsing forms of multiple sclerosis.
66% OF PATIENTS FROM ULTIMATE I AND II WERE TREATMENT-NAÏVE3
A post hoc analysis of pooled data from Phase 3 ULTIMATE I and II trials evaluated the efficacy of BRIUMVI vs teriflunomide in participants not receiving an approved DMT in the five years prior to study enrollment (treatment-naïve population) and in a subset of treatment-naïve participants with symptom onset ≤3 years prior to study enrollment (early treatment subpopulation) at Week 96.3
Limitations: The post hoc nature of the analyses; the definition of the treatment-naïve group as participants who had received no prior approved DMT within 5 years before enrollment; and the possibility that participants enrolled in the controlled trials may differ from the real-world population.

15% OF PATIENTS FROM ULTIMATE I AND II HAD HIGHLY ACTIVE DISEASE4
A post hoc analysis of pooled data from Phase 3 ULTIMATE I and II trials evaluated the efficacy of BRIUMVI vs teriflunomide in people with highly active relapsing multiple sclerosis at baseline.
Highly active disease in this analysis was defined as4:
Limitations: Post hoc pooled analyses of ULTIMATE I and II (Phase 3) evaluated outcomes at Weeks 12 and 96 in participants with highly active disease. Limitations include the post hoc design, small sample size, and potential differences from real-world populations. Conclusions about safety or efficacy cannot be drawn based on these data.

>85% OF ULTIMATE I AND II PARTICIPANTS ENTERED THE OLE TRIAL5
Patients enrolled in ULTIMATE I and II either continued BRIUMVI treatment or switched from teriflunomide to BRIUMVI.
The OLE trial was designed to evaluate the long-term efficacy and safety of BRIUMVI and lacked a control group; results of the OLE trial should be interpreted with caution.
DBP, double-blind period; OLE, open-label extension.
